Despite the fact that my brother and I have inherited many fine old lamps, which are stuffed into closets and dark corners throughout the house, the one on his nightstand is just such a size and shape to be perfectly suited for his needs. However, the lamp was beginning to malfunction, requiring certain obscure motions to be made when interacting with it lest it turn itself off. Being unable to continue in such a fashion any longer and despairing of the necessity of using another less perfect lamp from our collection, my brother asked me yesterday for advice on local purveyors of lighting appliances who might be able to supply a similarly shaped lamp for his use. While I was able to name a suggestion, at the same time I was already disassembling the problematic piece and examining the defect socket. Surely, as it easy as it was to take apart, it must likewise be trouble free to reassemble with a working socket and contrary to the laws of narrativium, it was in fact so. The light is once again in fine working condition and currently in service even as I type.

Today's bathroom scale reading says I've lost 10 pounds since taking up an exercise and healthier diet routine.

This is a petty accomplishment because that took two months and I still have 30 pounds to go.

My accounting software was showing a balance that was way off what the bank was telling me. I downloaded a csv file of the account and went through it line by line. Found the two errors that caused it.:pleased:

So just fyi, the work is actually pretty simple. There are these magical transporters called PTS phosophotransferase system. It brings sugar into the cell for free by tricking the universe. So things that can move in and out of the cell move by diffusion, basically the concentration on both sides achieves balance through the wonders of stats and brownian motion (little shit just jiggles). Cells generally have higher concentrations of sugar inside than found in their environment, but the pts tags the sugar with a phosphate, so instead of being glucose, now its glucose-6-phosphate, and it wont diffuse back out of hte cell. Later depending on the pathway, the cell will recoup that phosphate of energy. So I have always had a soft spot on my heart for pts.

There are different kinds of these pts, the one we worked with is in the mannose family (mannose is a kind of sugar). A permease is a protein that acts as a channel in the membrane letting a molecule in.

The organism we worked with was Salmonella, but don't worry it was totally one of the more pathogenic salmonella strains 14028s. :)

So with avoiding a true teal deer explosion of text, we found that these genes found in an operon (a group of genes transcribed together which is a cool prokaryote trick that we don't have), let in an aminosugar (which is an aminoacid bonded to a sugar), and were needed for the cell to use the aminosugar.

But yeah, I have been living a life of leisure here in California and loving it. Helping to produce this work fulfills a life goal. I am going to apply to a local masters program but even if I don't science more in the future, I am fucking published.

That would be an application, for that you would need an engineer, I'm just a poor country microbiologist :)

Seriously, conceivably this is knowledge that could effect dealing with salmonella or similar transporters in other species. Salmonella is a leading cause of food borne illness and pts are found in many, many organisms.

I laid some sod this weekend. It's just as filthy as it sounds. :bunnythrust:

We hired a guy to build a step and put in pavers, as well as prepare a 125-square-foot space for a small lawn in back. We'd torn out the lawn that was back there in a different spot but never grew properly 'cause it gets too much shade. The new lawn space is where the hot tub used to be. Our guy put in the bend-a-board and sprinklers, and filled in the spot with dirt, so all I had to do was add the lawn soil and sod. The sod rolls were somewhat heavy, although a couple of them were wet, which makes them like 3x heavier. I lugged all that shit myself. :muscles: There is something extremely satisfying about going from mud to solid grass in one afternoon.

Ms. Chickenbunny is quite enamored of it, which was the plan. Even though it's not the closest pooping spot to her doggie door, the very first morning after installation, she started going straight to the grass to do her business. Quite a relief after a week of her tracking goddamned mud into the house every day.

Oh, and we got the kitchen redone with granite this weekend, as well. I didn't do that myself (like I'd be remotely competent to do any part of that), but I did clean the 500 layers of dust that the work deposited on the floors. Just in time for a dinner party at our house, at which I attempted to stay awake and mostly succeeded.

I was sitting at my computer this afternoon. I took a mouthful of coffee, some of which went down the wrong way. I couldn't stifle a cough, but I managed to avoid spraying coffee all over my keyboard.

A word of advice, if you ever need to have any part of your home insulated, estimate the cost of doing it yourself, and then call the professionals for an estimate. The estimate of the professionals will probably be within a few dollars, and it will be done much more quickly, and you won't itch from doing it yourself.
